Protective Mechanism of Bismuth Nitrate against the Nephrotoxicity of Mercuric Chloride in Mice

Abstract
The protective effect and its mechanism of bismuth nitrate against nephrotoxicity of mercuric chloride were investigated in the kidney of mice pretreated with bismuth nitrate, buthionine sulfoximine (BSO, GSH synthesis inhibitor) and L-oxothiazolidine carboxylate(OTZ, GSH synthesis pretreated with bismuth nitrate, BSO and OTZ. Blood urea nitrogen(BUN) level of mice given mercuric chloride with bismuth nitrate significantly decreased in comparison with that of the mice given mercuric chloride alone, but renal protein-SH and nonprotein-SH concentration significantly increased in comparison with that of the mice given mercuric chloride alone. On the other hand, GSH synthesis inhibitor, BSO pretreatment abolished the protective effect of bismuth nitrate against nephrotoxicity of mercuric chloride. In addition, treatment with GSH synthesis stimulator, OTZ significantly reduced nephrotoxicity of mercuric chloride. The results of present study suggest that GSH plays an important role in the protective effect of bismuth nitrate against nephrotoxicity induced by mercuric chloride.
